Colony forming assays as well as live cell imaging in human breast cancer cell lines indicated that CHFR downregulation is associated with increased sensitivity to PLK1 inhibitors as well as paclitaxel in vitro that reflects premature mitotic exit and increased multinucleation...Moreover, there was a strong association between higher CHFR expression and lower relapse-free survival in estrogen receptor (ER) positive, HER2 negative cancers (HR 7.8, p = 0.024). These observations identify CHFR as a potential predictive marker of sensitivity in ER positive breast cancer that warrants further investigation and suggest that CHFR might also be associated with response to certain other mitotic inhibitors.

The present study demonstrates that elevated ctDNA methylation of the SOX11 gene is a biomarker for reduced RFS after curative-intent resection of LARC. Patients with high ctDNA methylation of the SOX11 gene may not be optimal candidates for LARC resection. A prospective study is necessary to further validate SOX11 ctDNA methylation as a biomarker for RFS of patients with LARC.

The most important is, the ectopic expression of CHFR significantly inhibited SGC-7901 cell-derived xenografts and obviously promoted lung metastasis of GC cell with NRF2, p-AKT and p-ERK increased. Taken together, our findings suggested that CHFR might take part in gastric cancer progression especially cancer metastasis by activating AKT and ERK via NRF2- ROS axis.

These cell lines included those not used in the screening, validating the usefulness of the combination. These results showed that gastric cancers with CHFR methylation were sensitive to KRAS inhibition, and that methylation synthetic lethality is a promising therapeutic strategy.
Likewise, no association between CHFR expression and paclitaxel sensitivity was observed in ovarian cancer PDXs treated with paclitaxel monotherapy. Accordingly, differences in CHFR expression are unlikely to play a major role in paclitaxel sensitivity of high grade serous ovarian cancer.

In contrast, no significant differences were found between different methylation patterns for CHFR and the response to taxane-based neoadjuvant treatment. These results suggest the potential role of the higher expression of CHFR and the methylation pattern of its promoter as potential prognostic biomarkers in PDAC, thus warranting further comprehensive studies to extend and confirm our preliminary findings.
