IRX3-CDK14 axis promotes glioblastoma progression by regulating LRP6-mediated canonical Wnt/β-catenin pathway. (PubMed, Cell Death Dis)
In addition, we identify LRP6 (LDL receptor-related protein 6) as a crucial regulatory factor in maintaining IRX3-mediated stabilization of β-catenin. Our results demonstrate that IRX3 serves as a promising biomarker for patients with GBM, and targeting the IRX3-CDK14-LRP6 axis may represent a viable treatment approach for GBM.

Targeting PABPC1: A Therapeutic Strategy of Natural Ganoderma Meroterpenoid LZ22 against Triple-Negative Breast Cancer Proliferation. (PubMed, Pharmacol Res)
Furthermore, through structural optimization of LZ22 guided by molecular docking, we identified a derivative (LZ22-4) with enhanced anti-tumor proliferative activity. These findings not only identify PABPC1 as a valuable therapeutic target for TNBC but also provide new opportunities and novel insights for advancing anti-TNBC drug development with LZ22 as a potential drug leading compound.

5-Hydroxytryptamine promotes non-small cell lung cancer metastasis via the SNRPG/WT1/CDK14 Axis. (PubMed, Mol Biomed)
Loss of SNRPG led to WT1 accumulation, which in turn bound to the CDK14 promoter and activated transcription, ultimately driving NSCLC metastasis. These findings collectively suggest that the 5-HT/SNRPG/WT1/CDK14 axis plays a pivotal role in promoting NSCLC metastasis and highlight 5-HT and SNRPG as potential therapeutic targets for NSCLC metastasis.

Cancer type and gene signatures associated with breakthrough infections following COVID-19 mRNA vaccination. (PubMed, NPJ Vaccines)
We used epidemiological data from 21195 patients with cancer and 180407 matched controls, including in-depth analyses in 216 cancer patients, to discover clinical and molecular determinants that predispose cancer patients to breakthrough infections. Patients with B cell malignancies, with differential expression of CD24, CDK14 and PLEKHG1, were most susceptible to breakthrough infections, suggesting that these patients may require more booster immunisations to ameliorate cellular responses and immune protection against COVID-19.

CDK14 regulates the development and repair of lung. (PubMed, Cell Death Discov)
Here, we found that genetic ablation of Cdk14 influenced pulmonary vascular endothelial cells and alveolar epithelial cells during mice embryonic development as well as repair of lung after bleomycin or lipopolysaccharide induced injury...Disruption of Cdk14 interferes with IFN-γ induced lung repair in vivo, suggesting potential crosstalk of CDK14 signaling and IFN-γ pathway. Our work highlights the importance of Cdk14 in lung development and regenerative repair through an uncharacterized CDK14- IFN-γ signaling axis.

Methylation-modulated PFTK1 regulates gefitinib resistance via Wnt/β-catenin signaling in EGFR mutant non-small-cell lung cancer cells. (PubMed, Commun Biol)
Moreover, FMF-04-159-2, a specific covalent inhibitor of PFTK1, can reverse the effect of PFTK1 on gefitinib resistance in vitro and in vivo. Consequently, these findings shed new light on the mechanism underlying gefitinib resistance, and suggest PFTK1 as a target for gefitinib treatment in NSCLC.

In PCa lung metastases genes associated with immunogenic responses are upregulated while genes associated with epithelial-mesenchymal transition are down-regulated. This points to a more immunogenic phenotype of PCa lung metastases thus potentially making patients more susceptible to immunotherapeutic approaches.
