Generation of TCRγδ + T cells from human embryonic stem cells. (PubMed, Sci Rep)
RNA sequencing analysis revealed that the differentiation process of hESC-derived γδT cells was transcriptomically similar to those of in vivo courses. Therefore, our findings ultimately offer a novel technique to address the challenge of sourcing γδT cells for ACT therapies.

Myeloid sarcoma of the skull base: a brief review of the literature and a rare case confused radiologically with end plaque meningioma. (PubMed, Neurol Sci)
This case underscores the diagnostic challenge of skull-base MS due to radiological mimicry and highlights an unusual brachial plexus extension, broadening its clinical spectrum. biopsy, prompt systemic therapy, and multidisciplinary management are essential, particularly in the presence of a very high Ki-67.

Vaginal Myeloid Sarcoma: A Rare Extramedullary Presentation of a Myeloid Neoplasm. (PubMed, Cureus)
The patient was treated with 16 cycles of chemotherapy and underwent allogeneic bone marrow transplantation. This case illustrates the diagnostic challenge of vaginal myeloid sarcoma and emphasizes the role of imaging, immunohistochemistry, and multidisciplinary management in such rare presentations.

Peripheral blood plasmablasts, a potential confounder in chronic lymphocytic leukemia measureable residual disease analysis. (PubMed, Cytometry B Clin Cytom)
Clinical implementation of the CLL assay revealed 12 out of 77 (16%) CLL PB patient samples demonstrating a small population of plasmablasts. Plasmablasts normally exist in peripheral blood at levels detectable by flow cytometry MRD assays and may be a potential confounder in the identification of MRD in CLL.

IL-7 promotes the formation of DNA double strand breaks and DNA repair in murine pro-B cells. (PubMed, Front Immunol)
It is well established that IL-7 promotes the proliferation and survival of acute lymphoblastic leukemia (ALL) cells. Our data suggest that drugs targeting IL-7 could improve ALL therapeutic protocols.

CD169+ macrophages identify and eliminate tumor cells in colorectal cancer through CD169/CD43 interaction and FasL-driven apoptosis. (PubMed, Cell Rep)
This interaction, mediated by CD169/CD43 molecular engagements, leads to tumor cell apoptosis via high levels of factor-related apoptosis ligand (FasL). Our results not only deepen our understanding of the CRC tumor microenvironment but also open avenues for cancer immunotherapy targeting.

Surfaceome CRISPR activation screening uncovers ligands regulating tumor sensitivity to NK cell killing. (PubMed, bioRxiv)
Furthermore, we find that targeting CD43 on either NK cells or engineered T cells substantially enhances their cytotoxic activity against leukemia cell lines. These results establish gain-of-function screening as a powerful approach for discovering immunoregulatory surface proteins and identify multiple promising targets for enhancing NK cell-based cancer immunotherapies.

Subcutaneous Leukemia Cutis as the Initial Manifestation of CD23 Negative CLL/SLL in a Patient With Rheumatoid Arthritis on Chronic TNF-Alpha Inhibition. (PubMed, Case Rep Hematol)
While treatments for CLL/SLL-induced leukemia cutis vary, in this case, consolidative local radiation led to resolution of the remaining cutaneous lesion. Caution is advised when considering the use of TNF-alpha inhibitors in patients with a history of lymphoma.

Immunophenotypic, cytogenetic, and mutational features of chronic lymphocytic leukemia/small lymphocytic lymphoma with atypical immunophenotype. (PubMed, Cytometry B Clin Cytom)
In summary, approximately 20% of CLL cases show an atypical immunophenotype, and these cases have cytogenetic abnormalities and mutation profiles that differ in frequency from typical CLL cases. Recognition of the immunophenotype of atypical CLL can be helpful in the diagnosis and differential diagnosis in low-grade B-cell neoplasms.
