Development of DARPin T cell engagers for specific targeting of tumor-associated HLA/peptide complexes. (PubMed, iScience)
A cryo-EM structure of the ternary DARPin/HLA-A∗0201/NY-ESO1157-165 complex revealed a rigid, concave DARPin surface spanning the full length of the peptide-binding cleft, contacting both α-helices and the peptide. The present findings reveal promising immuno-oncotherapeutic approaches and demonstrate the feasibility of rapidly developing DARPins with high affinity and specificity for HLA/peptide targets, which can be readily combined with a new generation of anti-CD3e-specific DARPins.

Multiplex gene-editing strategy to engineer allogeneic EGFR-targeting CAR T-cells with improved efficacy against solid tumors. (PubMed, Nat Commun)
This combinatorial genetic disruption enhances CAR T cell effector function and anti-tumor efficacy leading to improved tumor elimination and survival in xenograft and humanized mouse solid tumor models. Our strategy confers CAR T cells resistance to multiple clinically relevant inhibitory signaling pathways that are amplified in hypoxic tumor areas and may improve the therapeutic potential of CAR T-cells against solid tumors.

In HER2-positive BC, a three-gene signature (IL2RG, CD3E, CD3G) was consistently prognostic (HR 0.41, 95% CI 0.24-0.71, P = 0.0011, GEO) across independent datasets. DNA methylation mediates the immunosuppressive tumour microenvironment in metastatic endometrial clear cell carcinoma. (PubMed, EBioMedicine)
This study illuminated the "epigenetic-immune axis" as a central regulatory mechanism driving ECCC metastasis. DNA methylation systematically silenced immune response genes by targeting ETS1-binding sites and TCR signalling components, thus reconstructing the immunosuppressive TME to facilitate metastasis. The development of the metastatic risk score model and identification of LCK as a potential therapeutic target provide valuable strategies for precision treatment decisions and advancing targeted epigenetic-immune therapies in ECCC.

Analysis of public data sets shows that most ORs are significantly downregulated in cancer, suggesting their grassroot tumor suppressors role when a network emerged as they teamed together against cancer.
