Strikingly, pharmacological blockade of CaMKIIδ using the small-molecule inhibitor hesperadin effectively eliminates AML cells in vitro and achieves sustained disease regression in AML xenograft mouse models. Our findings establish CaMKIIδ as a central regulator of AML cell survival and apoptosis, providing a preclinical rationale for targeting CaMKIIδ to overcome therapy resistance in AML.

Notably, hypophosphorylation at TPD52_S171/S176 was observed in hepatocellular and lung carcinomas, whereas hyperphosphorylation at TPD52L2_S166 prevailed in ovarian and pancreatic cancers, underscoring biomarker utility. Phosphorylation-driven interactomes emphasized roles in vesicular trafficking and oncogenesis This study catalogues the phosphorylation events and explores the potential of TPD52 family as a phosphoregulated hub in cancer biology, with CAMK2D as a potential therapeutic target.

Finally, we recapitulate the glioblastoma associated AMY1B and RN7SKP123-MTF2 chromosomal rearrangement using an extrachromosomal assay. Thus, the present study provides mechanistic insights into the generation of chromosomal aberrations associated with glioblastoma.

OSW-1 displayed strong synergy with 5-fluorouracil (5-FU), effectively eliminating RIP3-silenced CRC cells and tumors through the induction of both necroptosis and apoptosis...Furthermore, the combination increased immune infiltration in patient-derived primary CRC Air-Liquid Interface (ALI) organoid cultures with autologous immune tumor microenvironment. Collectively, our findings support a novel strategy to activate RIP3-independent necroptosis, providing a compelling rationale to boost the efficacy of chemotherapy via enhanced CRC cell killing and antitumor immunity.

RBFOX1 loss reverts cardiomyocytes to a less mature state with open chromatin and susceptibility to DNA damage, apoptosis or CIC, when treated with DNA intercalating or alkylating anticancer agents. These findings suggest cumulative inosine and hypoxanthine levels may be a biomarker predicting patient susceptibility to DNA damaging anti-cancer agents.

Furthermore, OSW-1-induced necroptosis is highly immunogenic and sensitizes syngeneic colorectal tumors to anti-PD-1 immunotherapy. Together, our results identified a novel RIP1/RIP3-independent necroptosis pathway underlying the extremely potent anticancer activity of OSW-1, which can be harnessed to develop new anticancer therapies by selectively stimulating antitumor immunity.

CAMK2D (Calcium/Calmodulin Dependent Protein Kinase II Delta) • RIPK1 (Receptor Interacting Serine/Threonine Kinase 1)
Sumoylated MZF1 can interact with proteins containing SUMO-interaction motifs (SIM) through SUMO-SIM interaction. Interactome analysis revealed that its NBs participate in the stress response (TPR and UBAP2L), protein folding (CALR and ANKRD40), transcription, post-translational modification (TRIM33, ACOT7, CAMK2D, and CAMK2G), and RNA binding (ALURBP and CPSF5).
This study also evaluates Tetrandrine (TET), a small molecule compound, as a potent CAMKIIδ inhibitor...Collectively, this study highlights how CAMKIIδ is critical in BCL progression. The results also pave the way for innovative therapeutic strategies for treating aggressive BCL.
More pre-clinical and clinical studies are needed to elucidate the precise mechanisms by which CEACAM1 RNA splicing may be exploited to develop targeted interventions towards novel therapeutic strategies.
Ecklonia kurome Okam (Kunbu) and Herba Ephedrae (Mahuang) were identified, and the effects of the active ingredients on AD were displayed. Finally, enrichment analysis of Kunbu and Mahuang suggested that they might act as therapeutic candidates for gender specificity of AD.
