In summary, paclitaxel induces transcriptomic and proteomic signatures of the neuronal stress response, neuroinflammation, nociception, and disturbed metabolism. These may explain, in part, the clinical phenotype of sensory loss, hypersensitivity, and neuropathic pain frequently observed in patients suffering from CIPN, but constitute pharmacologically addressable targets.

Loss- and gain-of-function approaches demonstrated that eIF2α-regulated, cap-independent translation is essential for recent consolidation and for the enduring stabilization of remote memory, whereas retrieval-induced destabilization engages a mechanistically distinct, eIF4E-dependent translational pathway required for reconsolidation. These findings identify the PL cortex as a dynamic node in which discrete modes of translational control govern the long-term persistence of emotional memories.

Non-neuronal cell types including astrocytes, oligodendrocytes, and vascular cells also exhibited region-specific translational changes in neurotransmitter transport, lipid synthesis, myelination, and stress response pathways, some of which co-varied with regional neuron state. Together, our study reveals brain-wide translation dysregulation as a critical mechanism underlying SCZ pathophysiology.

This study establishes a mitochondria-driven prognostic model for PDAC and identifies the CAMK2A-THEM4-AKT axis as a novel therapeutic target. To our knowledge, this is the first pancreatic cancer signature that investigates the synergistic effect of candidate genes, offering both prognostic and mechanistic insights into PDAC progression.

In vitro, PIK3CG, CAMK2A, NFKB1, PSMA4, and PSMB1 were upregulated in OC cell lines, and knockdown of PIK3CG notably suppressed the proliferative, migratory, and invasive capabilities of OC cells. The PRG model established in this study may contribute to the assessment of immunotherapeutic response and prognosis for OC patients.

Eleganolone, a diterpene isolated from seaweed Bifurcaria bifurcata (Phaeophyceae, Fucales), protects neuronal cells from oxidative stress-induced damage. (PubMed, Eur J Pharmacol)
Additionally, our data showed that eleganolone downregulated the expression of genes associated with apoptosis (BAX, BNIP3, p53, p38, APAF1), inflammation (NFKB1, TNF-α, IL-6, IL-1β), oxidative stress (HO-1) and neuronal development (CAMK2A, WNT5A, WNT7A) pathways in SH-SY5Y cells exposed to oxidative stressor tert-BOOH. These findings suggest that eleganolone offers neuroprotective potential in ameliorating diverse pathological aspects associated with oxidative stress.

Correction of dysregulated lipid metabolism normalizes gene expression in oligodendrocytes and prolongs lifespan in female poly-GA C9orf72 mice. (PubMed, Nat Commun)
Single nucleus transcriptomics indicated that CD primarily affected oligodendrocytes, significantly restored myelin gene expression, increased density of myelinated axons, inhibited the disease-associated oligodendrocyte response, and downregulated the lipid-associated genes Plin4 and ApoD. These results suggest that reducing excess free cholesterol in the CNS could be a viable ALS treatment strategy.
