Comparative mRNA expression analysis confirms that a short preoperative course of aromatase inhibitors induces a more potent and uniform molecular response, characterized by profound suppression of proliferation and complete inhibition of estrogen-dependent signaling. Tamoxifen therapy is also effective but results in less pronounced suppression of key targets and, crucially, may be accompanied by early activation of the MYC oncogene, a potential marker for resistance development.

Proteomic analysis of DEN-induced acute liver injury revealed alterations in key pathways in BAG3albKO mice livers, including inhibition of autophagy and increased liver necrosis. Collectively, these findings emphasize the complex role of BAG3 in HCC pathogenesis and indicate its participation in tumor onset and progression.

BAG6 promotes hepatocellular carcinoma growth via enhancing PLK1-mediated aerobic glycolysis. (PubMed, Int J Biol Macromol)
Mechanistically, BAG6 directly binds to PLK1 and suppresses its ubiquitination and degradation, and enhances PLK1-mediated Warburg effect, thereby promoting HCC growth. Overall, we identify BAG6 as an HCC progression driver via the PLK1-mediated Warburg effect, a potential therapeutic and early screening marker.

Targeting the BAG2/CHIP axis promotes gastric cancer apoptosis by blocking apoptosome assembly. (PubMed, Front Immunol)
Furthermore, we screened FIIN-2, an inhibitor of the BAG2 complex, which effectively halts the malignant development of GC triggered by reduced apoptosis by blocking BAG-CHIP binding. In conclusion, this study highlights BAG2's key role in regulating apoptosis and confirms FIIN-2's effectiveness in GC-targeted therapy.

Notably, we discovered that the natural compound sophoricoside (Sop) acts as a METTL21A inhibitor and exhibits potent anti-HCC activity.
