Mechanistic study of ANXA3-mediated endoplasmic reticulum stress promoting M1 macrophage polarization in pulmonary arterial hypertension based on bioinformatics and nine machine learning algorithms. (PubMed, Comput Biol Med)
This study demonstrates that ANXA3 regulates ERS to drive M1 macrophage polarization and inflammation, which subsequently promotes PASMC function and promotes PAH progression. ANXA3 may serve as a novel immunoinflammatory target and potential therapeutic candidate.

Shared diagnostic genes and potential mechanisms between asthma and lung cancer revealed by integrated transcriptomic analysis and machine learning. (PubMed, Eur J Transl Myol)
Utilizing machine learning-based screening, we identified three hub biomarkers: P2RY14, ANXA3, and SLIT2, which could serve as diagnostic tools for these diseases. In summary, our research provides invaluable insights into the shared mechanisms underlying asthma and lung cancer, and potential diagnostic biomarkers.

Assessing the role of Escherichia coli and Klebsiella pneumoniae in colorectal cancer oncogene expression: insights from microbial colonization phenotypes. (PubMed, Mol Biol Rep)
Our findings indicate that E. coli and K. pneumoniae may exert nuanced, tumor-specific effects on oncogenic pathways, potentially mediated more by chronic inflammation than direct gene dysregulation. Further studies employing single-cell RNA sequencing are warranted to elucidate these interactions and their translational potential for CRC diagnostics and therapeutics.

Also, ANXA3 silencing could reduce lung cancer tumorigenesis and enhance DDP sensitivity by decreasing ANXA4 expression in vivo. ANXA3, activated by FOXD1, might contribute to the DDP resistance of lung cancer via regulating ANXA4, providing new ideas for overcoming chemoresistance in lung cancer.

Discovery of a Novel 1,4-Benzodiazepine Derivative as a Highly Selective ANXA3 Degrader for the Treatment of Triple-Negative Breast Cancer. (PubMed, J Med Chem)
In particular, 18a5 showed more desirable ANXA3 selective degradation than (R)-SL18 and displayed excellent inhibitory effect in a TNBC tumor xenograft model (TGI = 96%). Collectively, 18a5 proved to be a promising lead compound to treat TNBC through the degradation of ANXA3.

In summary, the discovery of PANoptosis genes, ANXA3, S100A9, TXN, CLEC5A, and TMEM263 proved to be quite helpful in the early detection of pediatric septic shock patients. These early results, which need to be further confirmed in basic and clinical research, are extremely important for understanding immune cell infiltration in the pathophysiology of pediatric septic shock.
