AML with t(4; 12)(q12; p13): A Detailed Genomic and Transcriptomic Analysis Reveals Genomic Breakpoint Heterogeneity, Absence of Pdgfra Fusion Transcripts and Presence of Pdgfra Overexpression in a Subset of Cases (ASH 2023)
Analysis of AML with t(4; 12)(q12; p13) translocation by WGS and WTS provides detailed information that separates two subsets distinguished by distinct breakpoint cluster regions on 4q12 and PDGFRA gene expression. The clinical impact of increased PDGFRA gene expression on response to TKI and prognosis as well as the presence of additional fusion transcripts has to be evaluated in further studies.

ADAMTS3 restricts cancer invasion in models of early breast cancer progression through enhanced fibronectin degradation. (PubMed, Matrix Biol)
We show further that loss of ADAMTS3 enhances levels of fibronectin in the microenvironment, promoting invasion through canonical integrin α5β1 activation. Our data highlight a tumour suppressive role for ADAMTS3 in early stage breast cancer, and contribute to the growing evidence that proteases can restrain cancer progression.

Body composition and lung cancer-associated cachexia in TRACERx. (PubMed, Nat Med)
Primary non-small cell lung cancers from individuals who developed CAC were characterized by enrichment of inflammatory signaling and epithelial-mesenchymal transitional pathways, and differentially expressed genes upregulated in these tumors included cancer-testis antigen MAGEA6 and matrix metalloproteinases, such as ADAMTS3. In an exploratory proteomic analysis of circulating putative mediators of cachexia performed in a subset of 110 individuals from TRACERx, a significant association between circulating GDF15 and loss of body weight, skeletal muscle and adipose tissue was identified at relapse, supporting the potential therapeutic relevance of targeting GDF15 in the management of CAC.

The Potential of Extracellular Vesicle Derived microRNAs As a Biomarker in Acute Myeloid Leukemia (ASH 2022)
Conclusion Overall, the findings of this study revealed that EV-derived miRNAs may serve as biomarkers for risk stratification and prognosis in AML. However, further studies are required to validate these EV-derived miRNAs in other cohorts before their implementation in clinical practice.

When validated with two other independent cohorts, the 6-gene risk score models remain a significant predictor for OS. Investigating the common gene expression program is significant in that we may extrapolate the findings from adults to children and avoid unnecessary pediatric clinical trials.

In conclusion, adults with a high PRS, particularly those experiencing early menarche, may particularly benefit from not exceeding energy requirements and from regular physical exercise in order to prevent abdominal obesity. This finding can be applied to personalised nutrition advice to prevent abdominal obesity.

Construction of a solid Cox model for AML patients based on multiomics bioinformatic analysis. (PubMed, Front Oncol)
Using the LASSO model, we built a classifier based on 5 hub genes to predict the prognosis of AML (risk score = -0.086×ADAMTS3 + 0.180×CD52 + 0.472×CLCN5 - 0.356×HAL + 0.368×ICAM3). In summary, we constructed a prognostic model of AML using integrated multiomics bioinformatic analysis that could serve as a therapeutic classifier.
