Biomimetic core-shell breast cancer models using alginate, gelatin, and collagen I: simulating the tumor matrix for drug evaluation. (PubMed, Int J Biol Macromol)
Breast cancer cells proliferated in the core of all prototypes designed, forming spheroids and cell aggregates with a high resistance to doxorubicin. The addition of Collagen I to the developed model enabled the upregulation of malignancy markers (Col1A1, Ki67, FOXC2, SNAI1, NFKB1, WWTR1), invasion markers (WASL, ACTA1, MYO1E, TPM4, PODXL, ITGA2, ITGA5, MENA, EGFR, CDC42), and drug resistance markers (ABCG2, CYP1A1, BAX, HSP90AA1) occurring in vivo. The developed 3D in vitro model can clarify the contribution of the extracellular matrix to the tumor outcome and drug efficacy by replicating some key characteristics of breast tumors, establishing a novel tool for chemotherapeutic agents and drug screening.

High GLP1R gene copy numbers associated with microsatellite instability for multiple cancers and with better survival probabilities for glioblastoma. (PubMed, Cell Cycle)
Results also indicated that high microsatellite instability directly correlated with high CNs for most of the above indicated genes. These approaches to assessing tumor metabolism-related genes may lead to more accurate measures of patient risk and potential additional treatment options.

An Evaluation of Prognostic Factors in Cutaneous Squamous Cell Carcinoma: A Single-Center Study of 237 Japanese Cases. (PubMed, J Clin Med)
RNA sequencing revealed the significant amplification of ACTA1, which was confirmed by Western blotting and immunohistochemical staining. These results suggest that HS is a prognostic factor in cSCC and that LL-37 stimulation contributes to tumor progression, partly by enhancing multiple tumor growth factors.

Gastric adenocarcinoma with enteroblastic differentiation. (PubMed, Pathobiology)
GAED is a rare subtype of gastric adenocarcinomas, representing 0.3-5.4% of all cases and has an unclarified etiology. Our WES results identified new pathogenic and likely pathogenic mutations. From a differential diagnostic point of view, hepatoid adenocarcinoma, and the possibility of metastatic origin has to be excluded.

Integrating clinical and genomic landscape analysis of perineural invasion identify ACTA1 as an oncogene for oral squamous cell carcinoma. (PubMed, Front Cell Dev Biol)
Among them, Actin α1 (ACTA1) was significantly upregulated in PNI+ advanced OSCC with worse clinical outcome whose had relatively low ratio of CD3+CD8+ circulating cytotoxic T cells. PNI+ OSCC patients with upregulated of Actin α1 could benefit from cytotoxic T cell-mediated immunotherapy.

Our results identified several genes with high centrality in PPI network that contribute to progression of CRC. The fact that several of the identified genes have already been reported to be relevant to diagnosis and treatment of CRC, other highlighted genes with limited literature information may hold potential to be explored in the context of CRC biomarker and drug target discovery.

Our study is the first to perform comprehensive and transcriptome-wide maps to identify the potential roles played by m6A methylation in BE, which widely involved in oxidative stress. This foresees a guiding role in revealing the molecular mechanism of m6A-mediated genes that govern the pathogenesis and progression of BE and EAC.
