    ...we assess the response and resistance mechanisms to the natural product derivative saridegib in an aggressive Shh-driven mouse medulloblastoma model....Saridegib inhibited reporter activity at an IC50 of 9 nM in C3H10T1/2 cells transfected with WT SMO and also showed activity against the D473H SMO mutant at an IC50 of 244 nM. Saridegib induced greater than 95% inhibition at 1 μM concentration in D473H SMO mutant cells...
