Datopotamab deruxtecan (DS-1062a) is another TROP2 ADC being explored in NSCLC. RR 20-25% here and well tolerated but pneumonitis of interest across compounds. No clear correlation with response and TROP2 expression but more data needed, esp as potential negative predictor (@StephenVLiu)

#ESMO21 TROPION-PanTumor01 enrolled patients without TROP2 expression requirement. The NSCLC expansion was at doses of 4mg/kg, 6mg/kg, and 8mg/kg q3w. The 8mg/kg dose did not have as great a therapeutic window but was explored in this phase I study. #LCSM @myESMO (@StephenVLiu)

TROP2 overexpression is per se a negative prognostic factor in Bladder, breast and lung cancer, so it’s an unmet medical need for solid tumors. Activity was shown in trials regardless expression stratification. breast cancer seems to be more evolved in this research certainly (@ChristianRolfo)
