While L768S and V773L have similar colony number with K753E after being treated with trastuzumab and lapatinib, the drugs significantly decreased the number of colonies formed, and we conclude that L768S and V773L mutations were sensitive toward trastuzumab and lapatinib treatment. Meanwhile, neither drug was able to significantly reduce the number of colonies formed by MCF10A cells with HER2 K753E mutation, indicating that K753E was resistant to these drugs.
